1.0 mL f/2 Trace Metal Solution (see recipe below)
0.5 mL f/2 Vitamin Solution (see recipe below)
Make final volume up to 1.0 L with filtered tertiary distilled water.
Note: f/2 Medium contains extensive silica precipitate and should be used only
when growing diatoms. For other algal groups use f/2-Si Medium (see below).
F/2 Vitamin Solution
(Guillard & Ryther 1962, Guillard 1975)
To 950 mL dH2O add:
Quantity Compound Stock Solution
1.0 mL Vitamin B12 (Cyanocobalamin) 1.0 g/L dH2O
10.0 mL Biotin 0.1 g/L dH2O
200.0 mg Thiamine HCl - Make final volume up to 1.0 L with dH2O.
Filter sterilize into plastic vials and store in refrigerator.

PES = Provasoli´s enriched Seawater Medium
General purpose marine medium for xenic cultures.
Preparation: add 1 tube (20 mL) of ES-enrichment to 1000 mL of pasteurized, filtered tertiary distilled water.
For ES-enrichment solution,
add the following to 100 ml glass-distilled water:
in 100 ml glass-distilled water
NaNO3 350 mg
Na2glycerophosphate .5 H2O 50 mg
Fe-solution 25 mL
PII metal-solution 25 mL
vitamin B12 10 µg
thiamine 0.5 mg
biotin 5 µg
Tris buffer (Sigma Co.) 500 mg
* Adjust to pH 7.8, dispense in tubes (20 mL/tube) and autoclave. Store at 10° C.
Fe-solution:
Dissolve 351 mg of Fe(NH4)2(SO4)2 . 6H2O and 300 mg of Na2EDTA in 500 mL of de-ionized or distilled water
PII metal solution:
in 100 ml glass-distilled water
Na2EDTA 100 mg
H3BO3 114 mg
FeCl3 6H2O 4.9 mg
MnSO4 H2O 16.4 mg
ZnSO4.7 H2O 2.2 mg
CoSO4. 7 H2O 0.48 mg
